An instrument known as a spectrophotometer measures light absorbance. In a spectrophotometer, light of a particular wavelength is passed through a sample, consisting of a solution in a transparent holder (a test tube or cuvette). The intensity of the light going into the sample (incident light) is compared with the intensity of light coming out of the other side, after passing through the solution (transmitted light). The ratio of these two quantities is used to calculate the absorbance of the solution. An absorbance of 0 means that no light of that wavelength is absorbed. The absorbance increases as more light is absorbed. If 90% of the light is absorbed, the absorbance value is 1.0, and if 99% of the light is absorbed, the absorbance value is 2.0.

The cupric tartrate starting reagent shows absorbance itself at 530 nm. This is corrected for with the reagent blank (the tube with reagent but no protein). However, at high protein concentrations, enough of the cupric tartrate many be used in the reaction to reduce the reagent blank absorbance; this will show up as a standard curve, which falls below linearity at high protein concentrations.
