Is it wise to run disk cleanup and disk defrag at the same time?

No... disk clean up should be run first. This way you remove junk files and then defrag has less to actually defrag. Additionally running any program that modifies the contents of your hard drive while you're in the middle of defragging can cause defrag to have to start all over again thereby making the defrag take much longer than necessary.

If i do a disk cleanup, will it erase all my files?

No it doesn't erase all ur file. its actually.......
• Remove temporary Internet files.
• Remove downloaded program files. For example, ActiveX controls and Java applets that are downloaded from the Internet.
• Empty the Recycle Bin.
• Remove Windows temporary files.
• Remove optional Windows components that you are not using.
• Remove installed programs that you no longer use.

How to undelete files under Windows Vista?

I accidentally deleted some important files from Recycle bin when running a disk cleanup utility. This happened recently so the files are hopefully not overwritten yet, however I can't find a way to recover them. One particularly is a large file (1.8 GB) so I'm not sure system restore would do the job.
Is there some hidden command in Vista or a FREE software that can do it?

NOTE: I tried some so-called free software, but typically they have a file size limit of 50KB or so unless you're willing to buy it.

The Windows.old folder gives you an extra layer of protection when you reinstall Windows.
