I'm going to assume you are not an organic chemist, and you do not have access to an organic chemistry lab.  I'm also going to assume you want to end up with something that is food safe.  If you want to extract curcumin from turmeric, you can soak ground turmeric in 99% ethanol for a day or two.  Filter the solids away with a coffee filter. You should end up with a nice amber colored ethanol solution.  This solution will contain curcumin.  Unfortunately, it will also contain other molecules from turmeric that are soluble in ethanol.  If you evaporate away all the ethanol (let it sit open in a pan until evaporates away and then bake it at 200F until it looks dry), you will probably get a gummy mess, but it will be a gummy mess that is more enriched in curcumin than the straight turmeric.To get pure curcumin you probably have to use chromatography (Chromatography), which is not something you can really do at home.How do you find the concentration of curcumin in a solution:To try and find how much curcumin is in a particular solution, the most straightforward method is to use HPLC.  Again, this isn't something you would do at home.  You would have to send it off to an analytical lab for analysis.  =However, if you want to estimate how much curcumin is in solution, you could purchase pure curcumin from a health store and make several solutions of it at different concentrations.  Then you could compare the color of your unknown to the color of the solutions at different concentrations.  Of course, this is prone to interferences from other colored species so it can be very inaccurate.
