You can only delete photos from Camera Roll library on your iPhone.
Since you have synced photos from your computer, it will stored separately in an album. You cannot delete this album through your iPhone.

To delete this album, you will have to connnect your iPhone to computer and open iTunes. Now go to Photos tab and deselect previously selected checkbox and click Apply button. The album will automatically delete from your iPhone.

Syncing new folders also deletes previous synced items. So whatever you need sync it all at once.

Photo Library contains a collection of all your imported photos from iTunes on your computer.
Hence, these photo cannot be deleted directly from iPhone.

Apple does not allow any changes to be made to imported photos.

Thus, to delete these photos, simply connect your iPhone to computer and launch iTunes.
Here go to Library and click on iPhone 5 tab. Next go to Photos tab and uncheck the box for "Sync Photos from" and tap on Apply button.

Now all your Photo Library photos will automatically be removed from iPhone 5.
