Finally there are right answers and wrong answers. There are quite a few questions which do not have a wrong answer but then most of them do. This is the answer "to". You reply or answer 'to' a question, or you address something using 'to'.You use 'of' in the descriptive sense: the Duke of Wellington, or in reference to something: he spoke of the novel, poignantly. Etc.

"Who knows the answer?" is correct. By the way, 'does' is not part of the verb 'to be'.
